Determination of Biogenic Amines in Paralichthys olivaceus by High Performance Liquid Chromatography

摘要 采用高效液相色谱(HPLC)法测定了牙鲆体内的多种生物胺。以邻苯二甲醛(OPA)为柱后荧光衍生剂,通过Capcell PakMG C18色谱柱的梯度洗脱,将酪胺、腐胺、尸胺、组胺、胍丁胺、亚精胺和精胺等7种生物胺进行良好分离。测试结果表明,7种生物胺在0.05~30 mg/L的范围内线性良好,相关系数都在0.999以上(n=6),加标平均回收率为88.8%~106.9%,相对标准偏差(RSD)为3.0%~9.3%,最低检出限为1.5μg/L。结果表明,该法快速、灵敏、重现性好,能够满足牙鲆中生物胺的检测需要。 The method for determination of biogenic amines in Paralichthys olivaceus by reversed-phase high performance liquid chromatography and post-column derivatization with o-Phthalaldehyde (OPA) was developed in this paper. Seven amines (tyramine, putrescine, cadaverine, histamine, agmatine, sperrnidine and spermine) were well separated with this method. And the seven amines had a good linearity in the range from 0.05 to 30 mg/L, with the correlation coefficients being above 0.999 (n=6). The average recoveries and the relative standard deviations of the seven amines in spiked sample were of 88.8%-106.9% and 3.0%-9.3%, respectively. The detection limits of all amines were 1.5μg/L. In conclusion, this method was rapid and sensitive, with high recovery and fine reproducibility, which can be used to detect biogenic amines in Paralichthys olivaceus.
