摘要
【目的]探讨兔肝缺血再灌后小肠超微结构、细胞因子、氧自由基的变化及奥曲肽的保护作用。【方法】采用Pring’s兔肝脏再灌注模型,将24只新西兰大白兔随机分为三组:A组(假手术组)、B组(肝脏缺血再灌注生理盐水组)、C组(奥曲肽预适应组)。三组动物再灌240min试验结束时取小肠组织行电镜检查超微结构的改变,同时取小肠黏膜纽织检测各纽兔小肠黏膜中肿瘤坏死因子a(TNF-a)、白细胞介素-1β(IL-1β)水平,以及其中的超氧化物歧化酶(SOD)和丙二醛(MDA)的含量。【结果】①C组脑组织超微结构病理改变较B组明显较轻。②奥曲肽处理组小肠组织中TNF-a、IL-1β、MDA水平显著低于缺血再灌注组(P〈0.05),而SOD水平显著高于缺血再灌注组(P〈0.05)。【结论】①奥曲肽预处理后明显减轻兔肝缺血再灌注损伤引起的小肠组织超微结构损害。②奥曲肽预处理后对兔肝缺血再灌注引起的小肠组织损伤起到保护作用,该作用主要是通过抑制细胞因子的表达和清除氧自由基。
[Objective] To explore the changes of ultrastructure, cytokines and oxygen free radicals in the intestine after hepatic ischemia-reperfusion in rabbits and the protective effect of octreotide. [Methods]Pringle' s rabbit hepatic ischemia-reperfusion models were established. Twenty four adult New Zealand rabbits were randomly divided into sham operation group (group A), hepatic isehemia-normal-saline-reperfusion group (group B) and octreotide preconditioning group(group C). The changes of ultrastructure in the intestine were observed by transmission electron microscopy at 240min(T8) after reperfusion. The levels of tumor necrosis factor-alpha(TNF-a) and interleukin-lbeta (IL-1β) and the contents of superoxide dismutase(SOD) and ma- londialdehyde (MDA) in intestine tissue of each group were measured. [Results] The pathological change of intestine ultrastructure of group C was slighter than that of group B. The levels of TNF-a, IL-1β and MDA in group C were lower than those in group B( P d0.05). The level of SOD in group C was higher than that in group B ( P d0.05). [Conclusion] Octreotide preconditioning can relieve the injury of intestinal ultrastructure after hepatic ischemia-reperfusion in rabbits. Octreotide preconditioning can protect intestine tissue injury after hepatic ischemia-reperfusion in rabbits. The protective effect of octreotide is exerted mainly by inhibiting the expression of cytokines and scavenging oxygen free radicals.
