高效毛细管电泳法快速测定内江猪血清头孢唑啉钠 被引量:2

摘要 报道用高效毛细管电泳技术快速测定内江猪血清头孢唑啉钠(cefazolinsodium,CEZ)的方法。血清用乙腈去除蛋白质,经离心后上清直接进样,毛细管内径75μm,有效长度40cm,在50mmol/L碳酸钠/碳酸氢钠缓冲液(pH10)中,电压20kV分离5分钟,254nm紫外检测。此方法操作简单,测定迅速,线性范围为5~320μg/ml,日内相对标准偏差为1.7%,日间相对标准偏差为8.0%,平均加样回收率为107.9%,最低检测限为1.5μg/ml,定量灵敏度为3.6μg/ml。按16mgCEZ/kg对6头成年内江猪作静脉注射,测定结果经药动学软件分析得到药代动力学参数,结果表明内江猪对CEZ的代谢为二室开放模型,清除率为8.43ml/(kg·min)。 A rapid high performance capillary electrophoretic method was established to assay cefazolin in porcine blood serum. Porcine serum was deproteined by 60% acetonitrile and superlants were injected with 10s pressure load. The final assay condition was: 40cm75m silica capillary, 50mmol/L bicarbonate buffer at pH10, 20kV for 5min, 254 nm UV detection. The assay was linear within the range of 5320g/ml, the intraday and interday relative standard deviations ranged from 1.7% to 8.0%. Analytical recovery of cefazolin was 107.9%, the limit of detection was 1.5g/ml, the limit of quantitation was 3.6g/ml. To investigate the pharmacokinetic profile of cefazolin in pig, six adult pigs were injected with 16mg/kg cefazolin and all data was analyzed by pharmacokinetic software 3P87. Cefazolin metabolism in adult pigs appeared to be a typical twocompartment model with a clearance of 8.43ml/(kgmin).
