摘要
目的探讨体外培养大鼠骨髓间充质干细胞(BMSCs)的一些生物学特性。方法通过密度梯度离心和贴壁培养相结合的方法,分离纯化BMSCs。经体外培养扩增,观察其生物学特性。结果分离获得了高纯度贴壁生长的BM-SCs,BMSCs原代培养呈均匀分布的集落样生长,呈梭形,细胞传代稳定;在体外培养条件下BMSCs具有较强的增殖能力,并能被诱导向脂肪细胞和成骨细胞分化。结论建立了BMSCs体外分离、培养的体系,探讨了BMSCs的生物学特性,为利用BMSCs进行疾病的细胞治疗提供实验基础。
Objective To study on rat bone mesenchymal stem cells(BMSCs) biological character istics in vitro.Methods Isolated and purified BMSCs from bone marrow of rat by density gradient centrifugation and adhering to the culture plastic.After successive subculture and amplification,observing BMSCs character istics.Results The isolated BMSCs were mononuclear cells in bone marrow,their living behaviors was quite stable,and were induced to adipose cell and sclerotomal cell.Conclusion A method for isolation and purification in vitro of BMSCs from bone marrow has been established and BMSCs biological character had been learned.This provide examination foundation for cell therapy to disease.
