脐带间质干细胞多向分化潜能的鉴定

Multiple differentiation potential of human umbilical cord mesenchymal stem cells

摘要 目的:研究脐带间质干细胞的多向分化潜能,为心血管组织工程研究提供一个新的种子细胞来源。方法:采用胶原酶胰酶消化脐带组织,将获得的细胞传代培养,流式细胞仪鉴定细胞表面分子,再以不同方法使其向骨、脂肪、心肌和内皮细胞方向诱导,分别采用Von Kossa、油红O染色和免疫组化对分化的细胞进行鉴定。结果:脐带间质干细胞分别向成骨细胞和成脂肪细胞诱导培养后,Von Kossa染色和油红O染色阳性。在5-氮胞苷诱导后,心肌特异性抗体肌动蛋白α-actin和肌球蛋白myosin免疫组化染色阳性。在VEGF诱导后,细胞形成网格样结构,免疫荧光示CD31、CD34染色阳性。结论:脐带间质干细胞具有分化为骨、脂肪、心肌和内皮细胞的潜能,是非常有前途的心血管组织工程种子细胞来源。 Objective:To study the differentiation potential of human umbilical cord mesenchymal stem cells into osteogenic, adipogenic, myocardiogenie and endothelial cells. Methods:The human umbilical cord mesenehymal stem cells were cultured and differentiated in the osteogenic, adipogenie, eardiomyogenic and endothelial medium. At the end of the culture, the cells were stained with Von Kossa or Oil red-O solution to reveal osteogenic and adipogenic differentiation. Immunocytochemistry and immunofluorescenee were performed to investigate differentiation of the cells to cardiomyocytes and endothelial cells. Results:The human umbilical cord mesenchymal stem cells were positive for Von Kossa staining and have the ability to accumulate different amounts of lipid vacuoles after cultured in osteogenic and adipogenic medium. Immunocytochemistry showed that differentiated cells were strongly stained with cardiac sarcomefic α-actin and cardiac myosin after 5-azacytidine treatment. Immunofluorescence studies confirmed their endothelial phenotype with expression of known endothelial cell markers including CD31 and CD34 after VEGF treatment. Conclusion: Our results demonstrated that human umbilical cord mesenchymal stem cells have the ability to differentiate into osteogenic, adipogenic, myocardiogenie and endothelial cells.
