香蕉枯萎病菌的分离鉴定与形态学研究 被引量:5

摘要 香蕉枯萎病是一种毁灭性真菌病害,对香蕉的种植和产业发展危害严重,其病原为尖孢镰刀菌古巴专化型(Fusarium oxysporum f.sp.cubense)。为了研究该病原菌的致病机理和香蕉的枯萎病抗性机理,笔者采用组织分离法,对分离纯化得到的候选真菌菌株进行了形态学和显微镜观察,采用CTAB法提取了真菌DNA,并对ITS序列进行了PCR扩增,通过ITS测序及生物信息学分析等手段,对该病原菌进行了分子鉴定。结果表明:分离得到的候选真菌为尖孢镰刀菌古巴专化型,是香蕉枯萎病的致病真菌。 Fusarium wilt of banana is a destructive fungus disease caused by Fusarium oxysporum f.sp.Cubense,and which is serious harm to banana planting and its industry development. In order to study the pathogenic mechanism of the disease and the resistance mechanism of banana,tissue isolation method was used to isolate and purify the candidate fungus strain,which were observed by morphological method; CTAB method was used to extract DNA of fungus,and ITS (Internal Transcribed Spacers) sequence was amplified and analyzed; the pathogenic fungus were identified by molecular method. The results indicated that the candidate strain was Fusarium oxysporum f.sp.Cubense.,and which was confirmed as pathogenic fungi of banana wilt disease.
