摘要
以美国杏李品种为试验材料,利用Prunus的SFB基因特异性引物,对7个杏李品种进行PCR扩增,并进行克隆、测序。采用生物信息学分析方法分离鉴定了‘恐龙蛋’等7个杏李品种的花粉SFB基因。PCR结果显示,‘风味皇后’、‘味帝’、‘味王’、‘风味玫瑰’、‘味厚’、‘味馨’、‘恐龙蛋’等7个品种均扩增出一条460 bp左右的片段;对PCR产物回收克隆后测序,并通过BLAST比对和生物信息学分析表明,‘风味皇后’、‘味王’、‘风味玫瑰’、‘味厚’、‘味帝’、‘恐龙蛋’等6个品种中克隆到了一个SFBh基因;‘风味皇后’、‘味王’、‘风味玫瑰’、‘味厚’、‘味馨’等5个品种中克隆到了一个SFBe基因;‘味帝’和‘恐龙蛋’中分离克隆了SFBb基因;‘味馨’中分离克隆了一个SFB2基因;故7个杏李品种的花粉SFB基因型鉴定为:‘风味皇后’、‘味王’、‘风味玫瑰’和‘味厚’等4个品种的花粉SFB基因型相同,为SFBeSFBh;‘味帝’和‘恐龙蛋’的花粉SFB基因型相同,为SFBbSFBh;味馨’的花粉SFB基因型为SFBeSFB2。
Introduction of American plums was used as the experimental materials.Adopting SFB specific primer in Prunus,PCR amplification,cloning,sequencing of 7 Prunus simonii were carried out,and bioinformatics analysis method was adopted for isolation and identification of the SFB genes of 7 Prunus simonii.The resutts of PCR amplification show that a common fragment of about 460 bp was generated in 'Fengweihuanghou','Weidi','Weiwang','Fengweimeigui','Weihou','Weixin' and 'Konglongdan';DNA sequence bioinformatic analysis and BLAST reveal that a SFBh gene was separated from 'Fengweihuanghou','Weiwang','Fengweimeigui','Weihou','Weixin','Konglongdan';a SFBe gene was separated from 'Fengweihuanghou','Weiwang','Fengweimeigui','Weihou','Weixin';a SFBb gene was separated from 'Weidi' and 'Konglongdan';a SFB2 gene was separated from 'Weixin'.The SFB-genotypes of 7 Prunus Simonii as follows:'Fengweihuanghou(SFBeSFBh)','Weiwang(SFBeSFBh)','Fengweimeigui(SFBeSFBh)','Weihou(SFBeSFBh)','Weidi(SFBbSFBh)','Konglongdan(SFBbSFBh)','Weixin(SFBeSFB2)'.
