奥沙利铂长循环脂质体对人结直肠癌SW480细胞凋亡影响 被引量:2

Oxaliplatin long-circulating liposomes improves apoptosis of human colorectal cancer SW480 cells

摘要 目的探讨奥沙利铂长循环脂质体对人结直肠癌SW480细胞的凋亡作用。方法采用荧光标记的长循环脂质体,奥沙利铂长循环脂质体和游离奥沙利铂分别处理人结直肠癌SW480细胞,流式细胞仪、荧光显微镜检测细胞对脂质体的摄取;MTT分析载药脂质体对细胞增殖影响;透射电镜观察细胞超微结构变化;流式细胞仪及TUNEL检测细胞凋亡;Western blot分析actived-caspase-3(P17)的表达。结果脂质体被细胞摄取,2 h的荧光强度为(197.72±8.92),24 h的荧光强度为(641.74±44.46);2.6 mmol/L的奥沙利铂长循环脂质体(含28 mg/L奥沙利铂)对细胞的活性影响及凋亡诱导作用大于28 mg/L游离奥沙利铂(P<0.01);actived-caspase-3蛋白的表达随细胞凋亡增加而上调(P<0.01)。结论奥沙利铂长循环脂质体对人结直肠癌SW480细胞有显著凋亡诱导作用。 Objective To investigate the effect of oxaliplatin long-circulating liposomes(PEG-liposomal L-oHP) on the apoptosis of human colorectal cancer SW480 cells.Methods Colorectal cancer SW480 cells were treated with fluorescent labeled PEG-liposomes,PEG-liposomal L-oHP and free L-oHP,respectively.Cellular uptake of fluorescent-labeled PEG-liposomes were detected by flow cyometry(FCM) and fluorscence microscopy.The cells proliferation were analyzed by MTT assay.Cellular ultrastructure was observed with transmission electron microscopy(TEM).Cell apoptosis was detected by FCM and TUNEL.Expression of activated-caspase-3(P17) was analyzed with Western blotting.Results When cellular uptake of liposomes at 2 h,the average fluorescence intensity was 197.72±8.92,and at 24 h,the intensity was 641.74±44.46.PEG-liposomal L-oHP of 2.6 mmol/L(containing 28 mg/L L-oHP) induced a stronger apoptotic response than 28 mg/L free L-oHP(P0.01).The protein expression of P17 was up-regulated with apoptosis being enhanced(P0.01).Conclusion PEG-liposomal L-oHP exerts an obvious effect in inducing apoptosis of human colorectal cancer SW480 cells.
