摘要
利用RAPD技术对小麦(黑麦)异源重组系异源2号组配的22个强优势杂交组合的亲本进行了遗传差异检测分析。结果表明,被测材料间RAPD标记多态性较高.61个随机引物中,37个引物(占60.65%)扩增产物具多态性,共扩增得到181条带。其中,105条带具多态性,占58.01%。每个引物可扩增出1~7条多态性带,平均可扩增2.8条多态性带。父本异源2号与其强优势组合母本间RAPD遗传距离较大,平均为0.43,聚类结果也显示其单独聚为一类。由此证实,异源2号与母本间确实在分子水平上存在较大遗传差异。RAPD遗传距离与亲本各性状表型差异相关均不显著,与F1各性状杂种优势间除抽穗期外相关也均不显著。据此认为,难以直接利用RAPD遗传距离预测杂交组合的杂种优势。
The parents of 22 strong heterotic combinations,' in-which Yiyuan 2, a wheat-rye recombinant was used as male parent, Were tested and analyzed by random amplified polymorphic DNA (RAPD) markers technology. The results showed that the polymorphisms between tested objects were very high. Of the 61 10-iner arbitrary primers tested, 37 of them (60. 65 % ) were found to amplify polymorphic products, A total of 181 products were amplified, of which 105 (58.01 % ) products were found to be polymorphic. The polymorphic products amplified by each primers ranged from 1 to 7, with an average of 2. 8. The genetic distances(GD) between the male parent Yiyuan 2 and the female parents of its strong heterotic crosses were relatively greater, with an average of 0. 43. The results of cluster analysis by using UPGMA method alsoshowed that Yiyuan 2 was classified as one group by itself. So it was proved that there reallyexisted greater genetic diversities between Yiyuan 2 and the female parents at molecular level. The correlation between GD based on RAPD markers and genotypic diversity of each character of parents was all not significant. GD was also not significantly correlated with the hcterosis of each character of FI hybrids, except for heading date. It was considered that GD based on RAPD markers was dilfficult to be used as a direct predictor of hcterosis.
