甘蔗原生质体培养──一个可再生植株的细胞系的特征 被引量:2

摘要 用甘蔗PC9102的培养细胞分离的原生质体实现了植株再生。重复实验也获得成功。这个品系的嫩叶能诱导出淡黄色、小粒状的愈伤组织。在悬浮培养中,这种愈伤组织较快地破碎出大小不一、粒状细胞团。小细胞团是由小而圆、具有丰富内含物的细胞彼此紧密结合而成,不见细胞间质。小细胞团能长大。用这种悬浮细胞培养物易酶解出原生质体。原生质体植板率高,能再生粒状、坚实的“愈伤组织”,最后分化出完整的植株。 Plant regeneration could be real lied by culturing the protoplast isolatedfrom ' typical embryogenic cells' established from calli derived from clone PC9102.Repetitive studies have been demonstrated. A light--yellow small granulan calli wasinduced from young leaves of this clone. These calli could be readily broken into asmall--granulated cell during suspension culture. The cell in the clumps were round,richly cytoplasmic and small. They aggregated closely together with littleintercellular space. Protoplast could be easily isolated from these cell clumps. Theprotoplast possessed a good plating efficiency and the potential for regenerating. agranulated compact ' calli' which could different late into organs and eventually growinto whole plants after further culture.
