摘要
目的观察束缚应激对大鼠主动脉5-HT1D和5-HT2A受体表达的影响及通心络和薤白提取物的干预作用。方法健康W istar大鼠60只,随机分为对照组、束缚应激组、薤白组、通心络组。观察大鼠一般状况、主动脉内皮细胞形态和结构,检测血中ET-1和NO水平,采用Real Time PCR和Western印迹检测主动脉5-HT1D和5-HT2A受体基因和蛋白表达。结果束缚应激所致抑郁状态可导致血管内皮结构和分泌功能损伤,同时5-HT1D mRNA和蛋白表达降低,5-HT2A mRNA和蛋白表达升高;通心络和薤白提取物可有效保护血管结构和功能,调节主动脉5-HT1D和5-HT2A受体基因和蛋白表达。结论通心络和薤白提取物可通过增强介导舒血管作用的5-HT1D mRNA和蛋白表达,抑制介导缩血管作用的5-HT2A mRNA和蛋白表达,从而对抑郁状态大鼠血管内皮功能发挥保护作用。
Objective To investigate the effect of restraint stress on the expression of 5-HT1D and 5-HT2A receptors in aorta of rats and the intervention effect of Tongxinluo and Allium extract.Methods Healthy male Wistar rats were randomly divided into control,restraint stress,Allium extract and Tongxinluo groups.The general condition,morphological changes of endothelium cell in artery,the levels of ET-1 and NO in blood were observed.The real-time reverse transcription PCR was used to detect the gene expressions of 5-HT1D and 5-HT2A receptors in artery.The Western blot was used to detect the protein expressions of 5-HT1D and 5-HT2A receptors in artery.Results The damages of the structure and function of endothelial cell were induced by restraint stress,the gene and protein expressions of 5-HT1D receptor were reduced,the gene and protein expression of 5-HT2A receptor were increased;Tongxinluo and Allium extract protected the structure and function of endothelial cell,regulated the gene expressions of 5-HT1D and 5-HT2A receptors in artery.Conclusions Tongxinluo and Allium extract can increase the gene and protein expressions of 5-HT1D receptor and decrease the gene and protein expressions of 5-HT2A receptor,produce the protective effects on endothelial function of depressive state rats.
