两种老年失眠证候大鼠模型的建立和评价 被引量:12

Establishment and evaluation of the two kinds of aging rat pathogeny models of insomnia

摘要 目的:建立并评价2种复合型老年失眠证候大鼠模型。方法:分别采用腹腔注射D-半乳糖+夹尾刺激+睡眠剥夺,腹腔注射D-半乳糖+环磷酰胺及氢化可的松+睡眠剥夺制作肝郁、血亏阴虚型老年失眠证候大鼠模型,观测大鼠体征、脑部自由基水平、血浆6-keto-PGF1α、TXB2及血象含量。结果:与空白对照组、单因素模型组比较,2种模型组的自由基水平均显著增高(P<0.05,P<0.01);与其他组比较,老年肝郁失眠证候模型大鼠TXB2含量显著降低,6-keto-PGF1α含量及6-keto-PGF1α/TXB2比值均显著升高(P<0.01),而老年血亏阴虚失眠证候模型大鼠全血血象显著降低(P<0.01)。结论:老年肝郁及血亏阴虚失眠证候大鼠模型均具有临床证候病理特征,是可行、可靠的复合型动物模型。 Objective: To establish and evaluate the two kinds of aging rat pathogeny models of insomnia.Methods: The insomnia rats model with the pathogeny of aging Liver-qi stagnationwas induced by D-galactose neck back subcutaneously,the stimulation of the rats tail leading to Liver-qi stagnation,sleep deprivation using modified multiple platform method(MMPM).The other insomnia rats model with the pathogeny of aging Blood-deficiency and Yin was induced by D-galactose neck back subcutaneously,intraperitoneal injection of cyclophosphamide and hydrocortisone,sleep deprivation using MMPM.Then observed the morphology,free radical in brain,and the contents of 6-keto-PGF1α,TXB2 in blood plasma.Whole blood hemogram.Results: compared with control group and single-factor model group,the two multiple factors model groups all showed the higher levels of free radical(P0.01).Compared with the other groups,multiple factors model group with aging Liver-qi stagnation showed the higher levels of TXB2,while the lower levels of 6-keto-PGF1α and the rate of 6-keto-PGF1α / TXB2 significantly(P0.01),the other multiple factors model group with blood-deficiency and Yin showed the lower levels of the whole blood hemogram significantly(P0.01).Conclusion: The two kinds of aging rat pathogeny models of insomnia that have pathological features of clinical syndromes,were viable,reliable animal model of the compound.
