摘要
目的探讨心脏手术过程中,人心肌组织缺血再灌注后Notch信号分子表达的变化。方法 20例接受体外循环心脏手术的患者,于体外循环前和心脏复跳后15 min后获取心肌样品,以Western Blotting检测Notch信号途径相关受体Notch1胞内区(NICD)和下游分子Hes1表达变化,分析NICD和Hes1在心脏缺血再灌注前后的变化。结果缺血再灌注后心肌组织NICD和Hes1的表达明显减弱,缺血再灌注前后NICD的western blotting信号强度与β-actin信号强度比值分别为:(4.28±0.45)和(1.55±0.36),P<0.01;Hes1信号强度与β-actin信号强度比值分别为:(4.61±0.52)和(1.31±0.32),P<0.01。结论缺血再灌注过程导致心肌组织Notch途径相关受体Notch1胞内区和下游分子Hes1表达下降,提示Notch信号途径可能在人心肌缺血再灌注损伤中发挥调控作用。
OBJECTIVE To investigate the changes of Notch signaling molecules expression after ischemic-reperfusion in human myocardium underwent cardiac surgery with cardiopulmonary bypass(CPB).METHODS Twenty patients underwent cardiac surgery with CPB were included in this study.Their myocardial samples were obtained before CPB and 15 min after reperfusion and rhythm restoration.The expression of Notch signaling pathway receptor Notch1 intracellular domain(NICD) and its downstream molecule Hes1 were detected by Western Blotting.The signal intensities were analyzed to compare the changes of NICD and Hes1 expression before and after ischemic-reperfusion.RESULTS The expression of NICD and Hes1 were declined significantly after ischemic-reperfusion,the Western blotting signal intensity ratio of NICD and β-actin before and after ischemic-reperfusion: 4.28±0.45 and 1.55±0.36,P〈0.01;the Western blotting signal intensity ratio of Hes1 and β-actin before and after ischemic-reperfusion: 4.61±0.52 and 1.31±0.32,P〈0.01.CONCLUSION Ischemic-reperfusion can decrease the expressions of NICD and Hes1 protein,which suggests Notch signaling pathway may play a regulatory role in myocardial ischemic-reperfusion injury.
