癌基因STAT3与结直肠腺瘤性息肉关系的研究 被引量:1

摘要 目的通过研究癌基因STAT3在结直肠腺瘤性息肉组织中的表达,探讨其在结直肠腺瘤性息肉发病机制中的作用。方法采用免疫组化MaxVisionTM法检测100例结直肠腺瘤性息肉、80例炎症性息肉和80例正常肠黏膜组织中STAT3蛋白的表达,结合临床资料进行分析。结果 STAT3蛋白的阳性反应主要定位于细胞质。STAT3蛋白在结直肠腺瘤性息肉、炎症性息肉和正常肠黏膜组织中的阳性率分别为59.00%、35.00%和31.25%。结直肠腺瘤性息肉组织中STAT3蛋白的表达显著高于炎症性息肉和正常肠黏膜组织(P<0.01)。在结直肠腺瘤性息肉组织中,STAT3蛋白的阳性率直径≥2cm的息肉高于直径<2cm的息肉,无蒂息肉高于长蒂息肉,绒毛状腺瘤高于管状腺瘤(P<0.05),其表达与年龄、性别无关(P>0.05)。结论 STAT3蛋白的高表达可能与结直肠腺瘤性息肉的发生、发展有关,那些STAT3蛋白阳性的组织细胞可能是潜在的癌前细胞。 Objective To By researching the expression of oncogene STAT3 in colorectal adenomatous polyp tissues,to explore the role it played in the genesis mechanism of colorectal adenomatous polyp.Methods The protein expressions of STAT3 were detected in 100 colorectal adenomatous polyp tissues,in 80 inflammatory polyp tissues and in 80 normal colorectal mucosal tissues by MaxVisionTM immunohistochemistry,and the relationship with clinical data were analyzed.Results The positive reaction of STAT3 protein was located mainly in cytoplasm.Positive expression rates of STAT3 protein in colorectal adenomatous polyp tissues,inflammatory polyp tissues and normal colorectal mucosa tissues were 59.00%,35.00% and 31.25%,respectively.The expression of STAT3 protein in colorectal adenomatous polyp tissues was higher than that in inflammatory polyp tissues and normal colorectal mucosal tissues(P0.01).Among the colorectal adenomatous polyp tissues,there was a higher expression rate of STAT3 protein for the polyps with diameter ≥2cm than that with diameter2cm,the expression rate of the polyps with free pedicle was higher than that with long pedicle,the expression rate of villous adenomas was higher than that of tubular adenomas(P0.05),no significant differences were found between STAT3 protein expression and such factors as age and gender(P0.05).Conclusion The overexpression of STAT3 protein may play an important role in the genesis and progression of colorectal adenomatous polyp,those STAT3-positive cells may be potential pre-cancerous cells.
