Exosomes致敏的树突状细胞介导的抗大肠癌LoVo细胞株的作用

摘要 目的分离LoVo细胞释放的exosomes,以其致敏树突状细胞(dendriticcell,DCs),观察其对细胞毒性T淋巴细胞(cytotoxicTlymphocytes,CTLs)的激活效应。方法离心超滤和蔗糖密度梯度离心法分离Lovo细胞释放的exosomes,常规方法从静脉血单个核细胞诱导DC,并分离T细胞,将LoVo细胞来源的exosomes冲击或未冲击的DCs与T细胞共培养。MTT比色法检测体外细胞毒活性。结果LoVo细胞分泌的exosome致敏的静脉血Des激活CTL的能力显著高于肿瘤冻融抗原致敏的DCs组,在效靶比为50:1时,两组CTLs对LoVo细胞的杀伤率为(67.9%VS38.7%,P〈0.05)。结论树突状细胞经来自LoVo细胞分泌的exosomes致敏后,能活化CTIs而杀伤肿瘤细胞。
