水杨酸钠对顺铂所致听性损害的拮抗作用研究

摘要 目的观察水杨酸钠(NaSA)对抗顺铂(CDDP)耳毒性的作用。方法选用健康、耳廓反射灵敏的白毛赤目豚鼠30只,随机分为两组:①CDDP+NaSA(100mg/kg)组和②CDDP+NS(对照组)。采用ABR、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及光镜技术,观察用药前后各指标的变化。结果①组的ABR阈值明显小于对照组(P<0.01)。①组血清和耳蜗组织中MDA含量较对照组明显降低(P<0.05,P<0.01),SOD活性较对照组明显升高(P<0.01)。耳蜗光镜标本显示,对照组外毛细胞受损程度较重,①组比对照组外毛细胞受损程度明显为轻。结论水杨酸钠对顺铂所致的耳毒性具有明显拮抗作用。 【Objective】To investigate the antagonist effects of sodium salicylate(NaSA) on the ototoxicity of cisplatin(CDDP) in guinea pigs.【Methods】Thirty clean adult albino guinea pigs were used in this experiment which were healthy and prier reflex was sensitive.They were randomly divided into 2 groups: ① CDDP+NaSA(100 mg/kg) group(experimental group) and ② control group.ABR,the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in serum and cochlea tissue,light microscope was used to evaluate their effects on hearing threshold,function of outer hair cells,serum and cochlea levels and morphology.【Results】The ABR threshold,serum and cochlea MDA levels in the experimental group were significantly lower than those in the control group(P 0.05).The activity of SOD in serum and cochlea tissue of the experimental group was significantly higher than that of the control group(P 0.05).The light microscope showed that the structural damage of hair cells was serious in the control group,while the structural damage of hair cells was milder in the experimental group than that in the control group.【Conclusion】Sodium salicylate can effectively reduce ototoxicity of cisplatin.
