摘要 目的建立高效液相色谱法同时测定小鼠血浆中大黄酸和大黄素的方法。方法血浆经酸化采用乙酸乙酯两次提取,色谱柱为Polaris-C18,流动相为甲醇︰0.1%磷酸(含0.05%三乙胺)(72︰28,v/v),流速为1.0mL/min;柱温45℃;检测波长为434nm。结果大黄酸和大黄素血浆浓度分别在0.0416~8.32μg/mL和0.044~8.80μg/mL范围内具有良好线性关系,方法回收率为96.4%~98.7%(大黄酸),96.8%~98.4%(大黄素),日内、日间相对标准差(RSD)均<6.0%。结论本方法简便、准确、灵敏、重现性好,可用于大黄酸和大黄素的血药浓度测定及药代动力学研究。 【Objective】To establish a simple method for simultaneous determination of Rhein and Emodin in mouse plasma by RP-HPLC.【Methods】Plasma sample was extracted twice with acetidin through acidification for RP-HPLC analysis.The chromatographic separation was performed on a Polaris-C18 column with a mobile phase consisting of methanol:0.1%phosphoric acid(including 0.05% TEA)(72:28,v/v) at a flow rate of 1.0 mL/min.The temperature was set at 45℃ and the detection wavelength was 434 nm.【Results】Rhein and Emodin were completely separated and no interference was found.The Rhein showed good linearity of the concentrations from 0.0416 to 8.32 μg/mL and the Emodin was from 0.044 to 8.80μg/mL in plasma and the recovery rate was 96.4 %-98.7 %(Rhein),96.8 %-98.4 %(Emodin);both the intra-day and inter-day RSD were less than 6.0 %.【Conclusion】 This method is simple,accurate,sensitive with good reproducibility,can be used for analysis of biological samples containing Rhein and Emodin and pharmacokinetic study.
