摘要
目的:动态观察肺纤维化大鼠的内皮素合成和分布,了解内皮素(Endothelin,ET)与特发性肺纤维化发生发展的关系。方法:采用放射免疫法和SP免疫组织化学方法,观察博莱霉素(BLM)组(n=25)和对照组(n=25)在1、3、7、14、28d时血浆ET浓度及肺组织ET的合成和分布情况。结果:BLM组大鼠血浆ET含量明显高于正常对照组(P<0.05),ET在支气管上皮细胞和炎性细胞表达增强(P<0.01)。结论:ET可能与肺纤维化的发病机理有密切关系,参与了肺纤维化的渗出和增生过程。
Objective: This paper was designed to observe the synthesis and distribution of
endothelin (ET) in rats with pulmonary fibrosis and to explore the relationship between ET and
the pathogenesis of idiopathic pulmonary fibrosis. Methods: We assigned fifty rats to two
groups each with twentyfive. Group normal rats received saline as the control, group fibrotic
rats received 0.2_0.3 ml blemony. On the lst, 3rd, 7,14 and 28th day, we measured their
plasma ET level by radioimmunoassay, and observed their ETlike immunoreactivity in lung
tissue morphologically by immunohistochemical staining. 5HZResults: The concentration of
plasma ET was significantly higher in pulmonary fibrosis rats than that in the control
(P<0.050.01). ET synthesis was mostly localized in bronchial epithelium and inflammatory
cells. In sharp contrast with group pneumocytes, there was little or no ET staining in normal
group. Conclusion: ET might be significantly related to the pathogenesis of pulmonary fibosis
in the roden model.
