摘要
目的:探讨人工关节磨损清洁钛微粒的大小及不同微粒-细胞比对巨噬细胞释放破骨性细胞因子:肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)、白细胞介素-6(IL-6)和前列腺素(PGE2)的影响。方法:清洁钛微粒按直径大小分为(0.35±0.09),(1.5±0.38),(6.25±2.13)μm组,分别以微粒-细胞比1:1,10:1,100:1,500:1,1000:1与巨噬细胞联合培养,用酶联免疫吸附法(ELISA)检测细胞培养上清液TNF-α、IL-1、IL-6、PGE2含量。结果:各组清洁钛微粒在微粒-细胞比10:1时,巨噬细胞释放TNF-α较对照组显著增高(P<0.05),500:1时释放TNF-α达到高峰,1000:1时分泌量下降。(0.35±0.09)μm清洁钛微粒组较(1.5±0.38)μm和(6.25±2.13)μm清洁钛微粒组,(1.50±0.38)μm清洁钛微粒组较(6.25±2.13)μm清洁钛微粒组,分别在微粒-细胞比10:1,100:1,500:1时,巨噬细胞释放更多TNF-α,具有统计学差异(P<0.05)。各组间IL-1、IL-6含量无显著性差异(P>0.05)。微粒-细胞比500:1,1000:1时,PGE2含量显著增高(P<0.01),巨噬细胞存活率显著下降(P<0.01)。结论:清洁磨损钛微粒大小及微粒-细胞比是决定磨损微粒生物细胞学反应的重要参量。磨损微粒越小,微粒-细胞比越高,生物细胞学毒性越大,能刺激巨噬细胞释放更多破骨性细胞因子TNF-α。清洁钛微粒本身并不能刺激巨噬细胞释放IL-1、IL-6、PGE2。
Objective:To study the effects of size-separated cleaned titanium particles and the ratio of titanium particles to macrophage cells on pro-inflammatory cytokines releasing in macrophage cells.Methods:Cleaned titanium particles were separated into three groups according to different diameter as(0.35±0.09),(1.50±0.38),and(6.25±2.13)μm.Macrophages were co-cultured in vitro with cleaned titanium particles at various particles to macrophage cells ratios(P-C ratio,1:1,10:1,100:1,500:1,and 1 000:1,respectively).The co-culture supernatants were assayed for tumor necrosis factor-alpha(TNF-α),interleukin-1(IL-1),interleukin-6(IL-6),and prostaglandin E2(PGE2)by enzyme linked immunosorbent assay(ELISA).Results:Compared with that in normal control group,macrophages in the other groups secreted more TNF-αsince at the P-C ratio of 10:1(P0.05),reached the peak at the ratio of 500:1,and decreased at the ratio of 1 000:1.When compared with the(1.50±0.38)μm and(6.25±2.13)μm groups respectively,macrophages co-cultured with(0.35±0.09)μm cleaned titanium particles significantly increased the release of TNF-αat a P-C ratio of 10:1,100:1,and 500:1respectively(all P 0.05).Macrophages co-cultured with(1.5±0.38)μm particles,versus(6.25±2.13)μm particles,secreted more TNF-αat P-C ratios of 10:1,100:1,and 500:1respectively(all P 0.05).It was found that cleaned titanium particles up to 1 000particles per cell did not stimulate IC-21macrophages to release IL-1and IL-6.Macrophages significantly increased the release of PGE2(P0.01)and the number of viable cells decreased at P-C number ratios of 500:1and 1 000:1(P0.01).Conclusion:The size and concentration of cleaned titanium particles are important parameters to decide the cytobiologic mode of macrophages.Smaller size and higher concentration of cleaned titanium particles,more TNF-αsecretion by macrophages.Cleaned titanium particles themselves can not stimulate macrophages to release IL-1,IL-6,and PGE2.
