摘要
以溴化乙锭(ethidium bromide,EB)为荧光探针,研究了更生霉素 D(actinomyin D,ACTD)与小牛胸腺DNA(calf thymus DNA,CT DNA,以下简称DNA)的作用机理.对荧光光谱、荧光偏振、Scatchard图、DNA热变性曲线、DNA盐效应曲线等研究表明,ACTD与DNA之间主要存在嵌入和沟槽两种作用.并证明ACTD与DNA磷酸基之间不存在静电作用.
The mechanism of the interaction between actionmycin D(ACTD) and calf thymus(CT) DNA has been studied by fluorescence method using ethidium bromide (EB) as a fluorescence probe. The results from varied experiments such as fluorescence spectra,fluorescence polarization, Scatchard curve, DNA heat denaturation, and DNA salt effect etc., support that intercalation and groove binding are the two major modes for the interaction ACTD and CT DNA. The absence of the electrostatic binding between the ACTD and the phosphate backbone of CT DNA has been proved.
