摘要
根据在巴西橡胶树幼态无性系与老态无性系胶乳之间差异表达的一个SSH片段序列信息设计引物,通过RACE技术获得了2个编码14-3-3蛋白的cDNA,命名为Hb14-3-3a和Hb14-3-3b。序列分析表明,Hb14-3-3a和Hb14-3-3b基因长度分别为1 154、1 050 bp,分别编码252、263个氨基酸,分子量为61.7 Ku和64.7 Ku,等电点为5.51和5.14。Hb14-3-3a/b基因在所检测的组织中均有表达,但Hb14-3-3a和Hb14-3-3b的表达模式不一样,Hb14-3-3a在树皮中表达量最高,而Hb14-3-3b在叶中表达量最高。
Two full-length cDNAs encoding 14-3-3 protein,designated Hb14-3-3a and Hb14-3-3b,were isolated from rubber tree based on the information about the sequence from a differentially expressed clone encoding 14-3-3 protein that was cloned recently by the subtractive hybridization between juvenile clone and the donor clones from rubber tree(Hevea brasiliensis).Sequence analysis revealed that the ORFs of Hb14-3-3a and Hb14-3-3b encode 252 and 263 amino acid residues with a total predicted molecular mass of 61.7 ku and 64.7 ku.The results of RT-PCR analysis indicated that the transcripts of Hb14-3-3a and Hb14-3-3b were differentially expressed in the flower,the leaves,the barks and the latex of Hevea brasiliensis.Hb14-3-3a transcripts accumulated at relatively high levels in the barks,while Hb14-3-3b transcripts accumulated at relatively high levels in the leaves.
