摘要
目的:研究消痰散结方对胃癌裸鼠原位移植瘤模型基因甲基化的影响,探讨消痰散结方抗癌作用机制。方法:培养胃癌细胞系MKN-45,制备胃癌裸鼠原位移植瘤模型,造模后分为模型对照组和中药组,分别给予生理盐水和消痰散结方水煎剂。称重法测定抑瘤率,RT-PCR法测定E-cad基因表达,Methylight法测定E-cad基因甲基化水平。结果:消痰散结方能抑制肿瘤生长以及转移,抑瘤率达45.17%。消痰散结方能逆转E-cad基因甲基化,增加E-cad mRNA表达,与模型对照组比较,差异有非常显著性意义(P<0.01)。结论:消痰散结方可以有效抑制胃癌细胞生长和转移,逆转E-cad基因甲基化可能是其作用机制之一。
Objective:To observe the effect of Xiaotan Sanjie Recipe(XSR) on E-cad methylation in nude mice with orthotopically transplanted human gastric cancer,and to explore the possible anti-cancer mechanism.Methods:Gastric cancer cell line MKN-45 was cultured,and the nude mice model of orthotopically transplanted human gastric cancer was established.The model nude mice were given normal saline(model group) and the decoction of XSR(XSR group),respectively.The anti-tumor rate was evaluated by weighting method,E-cad gene expression detected with RT-PCR,and E-cad methylation level examined with Methylight method.Results:XSR had an inhibitory effect on tumor growth and metastasis,the anti-tumor rate being 45.17%.XSR also counteracted the E-cad methylation,increased E-cad mRNA expression level,and the difference was significant as compared with the model group(P<0.01).Conclusion:XSR can effectively inhibit the growth and metastasis of gastric cancer cells,and counteract E-cad methylation,which may be one of its anti-cancer mechanisms.
