环磷酰胺对急性百草枯中毒小鼠肺损伤的保护作用 被引量:10

摘要 目的 探讨环磷酰胺对百草枯中毒致小鼠急性肺损伤的保护作用.方法 120只小鼠,随机分为4组,分别为空白对照(NS)组、百草枯对照(PQ)组、环磷酰胺对照(CP)组及环磷酰胺干预(T)组,NS对照组给予生理盐水×3 d,PQ组先给予生理盐水×2 d,第3天给予百草枯20 mg/kg染毒,CP组给予环磷酰胺80 mg/kg×2 d,第3天给予生理盐水,T组给予环磷酰胺80 mg/kg×2 d,第3天给予百草枯20 mg/kg染毒.染毒后第1、3、5、7天观察小鼠肺组织的病理变化并测定小鼠肺组织中的髓过氧化物酶(myeloperoxida, MPO)含量及血清中的肿瘤坏死因子(tumor necrosis factor-α, TNF-α)及白细胞介素-6(interleukin-6, IL-6)的含量并进行比较. 结果 PQ组染毒后血清IL-6含量第1天明显升高,后逐渐下降,多高于NS组,差异有统计学意义,血清TNF-α含量及肺组织MPO含量亦如此 经环磷酰胺预处理后的中毒小鼠,血清IL-6、肺组织MPO含量在第3、5天均明显低于PQ组,而第1、7天时差异无统计学意义,血清TNF-α含量第1、3天时显著低于PQ组,第5、7天差异无统计学意义.从肺组织病理来看,经环磷酰胺预处理的中毒小鼠肺组织的炎症细胞浸润较PQ组减少.结论 环磷酰对急性百草枯中毒所致肺损伤可能有一定的保护作用. Objective To study the protective effect of pretreatment of cyclophosphamide on lung injury of acute paraquat toxicity. Methods 120 second - grade mice were randomly divided into four groups. They were control group of normal saline, control group of paraquat, control group of cyclophosphamide and group of pretreatment of cyclophosphamide. The control group of normal saline were injected normal saline for 3 days. The control group of paraquat were injected normal saline for 2 days, then paraquat(20 mg/kg) in the third day. The control group of cyclophosphamide were injected cyclophosphamide(80 mg/kg) for 2 days, then normal saline in the third day. The group of pretreatment of cyclophosphamide were injected cyclophosphamide ( 80 mg/kg) for 2 days, then paraquat ( 20 mg/kg) in the third day. They were sacrificed in the first, third, fifth, seventh day after treatment to mensurate the content of myeloperoxida ( MPO), tumor necrosis factor - ct ( TNF - ct) and interleukin - 6 ( IL - 6), and observe the pathologic changing of lung tissue. Results The serum concentration of IL -6 of mice which were injected paraquat was obviously increased in the first day, and then decreased gradually. But they were higher than control group of normal saline respectively. The serum concentration of TNF - ct and lung tissue concentration of MPO were like this. Compared with PQ group, after giving the treatment of cyclophosphamide, the contents of IL - 6 and MPO decreased significantly in the third and fifth day. But they were not lower than PQ group respectively in other days. The serum concentration of TNF - α were lower than PQ group in the first and third day respectively but not in other days. Compared with PQ group, the infiltration of inflammatory cells reduced in the group of pretreatment of cyclophosphamide. Conclusion The pretreatment of cyclophosphamide maybe had some protective effect on lung injury of acute paraquat toxicity.
