摘要
以LacZ为报告基因,利用DNA重组技术构建了LacZ真核表达质粒pCDLacZ,pCDLacZ肌注小鼠后,X-Gal染色法及半乳糖苷酶定量分析结果表明,LacZ基因在小鼠胫前肌中得到表达,并在肌注后第7天左右达到高峰,表达的LacZ主要分布于胫前肌近膝关节肌腱结合处,提示肌肉接种基因疫苗应采用纵向,接种部位应接近肌腱结合处。
A karyotic expressing plasmid containing bgalactosidase(bGal) reporter gene was constructed, named pCDlaZ, after intramuscularily injecting plasmid pCDlaZ, the results of XGal staining and bGal quantitative analysis showed that a maximal level of bGal expression in mouse rectus femoris muscle reached about 7 days after injection of plasmid pCDlaZ, the bGal expression was located predominantly in the myotendinous junction areas.The results suggest that the intramuscular injection of genetic vaccine should be injected longitudinally and in the vicinity of myotendinous junction areas.\;
