摘要
目的:研究经前期综合征(PMS)肝气逆证边缘系统关键受体基因表达差异,揭示该病证基因水平部分发病机制。方法:选取高地位猕猴运用择时挤压造模法制备PMS模型,运用抑制性消减杂交技术构建PMS肝气逆证猕猴与正常猕猴边缘系统正向与反向差减cDNA文库,运用该文库制备表达谱基因芯片进行差异表达基因研究。结果:在猕猴模型边缘系统中中枢单胺类神经递质受体5-羟色胺1A受体(5-HT1AR)、5-羟色胺2A受体(5-HT2AR)mRNA表达均升高;γ-氨基丁酸A受体(GABAAR)mRNA表达降低;中枢甾体激素受体雌二醇α受体(EαR)、雌二醇β受体(EβR)mRNA表达降低;孕酮受体(PGR)mRNA表达升高。结论:上述6个受体基因与PMS肝气逆证中枢发病机制关系密切。
Objective:Studied on the differential expression of several important receptor genes in the limbic system tissue of PMS liver-qi invasion macaque model,reveal the gene level of some of the pathogenesis of the disease.Methods:This study selected high-status monkeys,used selecting time extrusion method to preparation of model,applied the technique of SSH in to acquire to construct limbic system positive and negative directions subtractive cDNA library,utilized the subtractive cDNA library and made macroarray gene chip to study different expression of genes.Results:In the limbic system tissue of PMS liver-qi invasion macaque model,nerve center monoamine neurotransmitters 5-HT1AR/5-HT2AR mRNA increase and CABAAR mRNA decreased;hormone EαR/E βR mRNA decreased,PGR mRNA increase.Conclusion:The six receptor genes are closely related with the Central pathogenesis of PMS.
