摘要
对小麦、油菜苗喷施浓度为0.2%水溶性壳聚糖后,叶片几丁质酶活性明显提高,油菜和小麦酶活性分别在处理后2d、6d时达最大值,均比相应对照提高2倍左右,且都表现为内切几丁质酶活性。叶片细胞间的几丁质酶比活力明显高于细胞内的酶比活力。壳聚糖诱导的油菜叶片几丁质酶在体外可抑制油菜菌核病菌生长,并能降解菌丝细胞壁。
Watersolublechitosan,apolymerof1,4Dglucosaminederivedfromcrabshellchitin,wasappliedtorape(Brassicanapus)andwinterwheat(Triticumaestivum)bymeansofleafspraying.Itwasfoundthatchitinase(EC3,2,1,14)activitieswasnearthreefoldinbothofleavesofwinterwheatandrapetreatedwith02%(W/V)chitosansolutiontothatinCK(sprayingTW80).Thepeakofchitinaseactivityinrapeandwinterwheatappearedatthe2nddayandthe6thdayrespectivelyafterchitosansolutionbeingsprayed.Chitoasninducedchitinaseshowedendochitinaseactivity.Theintercellularfluidofleaveswasextractedbythemethodofvacuuminfiltrationandcentrifugation.Resultsshowedthatchitinasehadmuchhigherspecificactivityinextractofintercellularfluidthanthatinleafhomogenates,andmostofthechitosaninducedchitinaselocatedintheextracellularspace.Chitinasefromrape leavescouldinhibitthegrowthofSclerotiniasclerotiorumandlysisthecellwallofSclerotiniasclerotiorumformingoligosaccharidesinvitro.Fromthatitcouldbeelucidatedthatchitosaninducedchitinasefunctionedasadefensiveenzymeagainstpathogenicfungi.
