摘要
目的:研究表皮调节素(epiregulin)在血小板源性生长因子-BB(platelet-derived growth factor-BB,PDGF-BB)促进大鼠血管平滑肌CS-54细胞株增殖中的作用。方法:针对大鼠表皮调节素基因的mRNA序列合成siRNA,采用脂质体转染法将其转染CS-54细胞;用蛋白质印迹检测PDGF-BB及siRNA转染对CS-54细胞表皮调节素蛋白表达的影响;用MTT法分析干扰表皮调节素表达对PDGF促进CS-54细胞增殖效应的影响。结果:PDGF-BB促进细胞增殖,效应具有浓度和时间依赖性,在50 ng/ml PDGF-BB作用24 h时其效果最为明显;PDGF-BB可以刺激CS-54细胞表皮调节素蛋白的表达;表皮调节素siRNA转染细胞,可使表皮调节素蛋白表达水平明显降低,并明显阻断PDGF-BB对CS-54细胞增殖的促进作用(P<0.05)。结论:PDGF-BB刺激CS-54细胞增殖的作用是通过诱导CS-54细胞表达表皮调节素蛋白实现的。
Objective: To study the role of epiregulin in rat vascular smooth muscle cell proliferation-stimulating effect of platelet-derived growth factor-BB(PDGF-BB).Methods: Epiregulin siRNA was synthesized according to mRNA sequence of epiregulin gene of rat and was transfected into CS-54.Western blotting was used to detect the expression change of epiregulin caused by PDGF-BB and epiregulin siRNA.The impact of PDGF-BB and epiregulin siRNA transfection on CS-54 proliferation was analyzed by MTT assay.Results: PDGF-BB could concentration and time-dependently promote CS-54 proliferation,most effective at 50 ng/ml PDGF-BB for 24 hours.PDGF-BB could stimulate CS-54 to produce epiregulin.Epiregulin siRNA not only decreased the level of epiregulin expression but also blocked proliferation-stimulating effect of PDGF-BB in CS-54 cells(P0.05,compared with control group).Conclusion: PDGF-BB stimulates proliferation via inducing the expression of epiregulin in CS-54 cells.
