摘要
以异硫氰酸酯(FITC)为指示剂,采用荧光法测定ε-聚赖氨酸(ε-PL)。体系的激发波长和发射波长分别为495nm和519nm。在2%乙醇介质中,ε-PL与FITC反应,使FITC发生荧光淬灭。ε-PL含量在0.1~0.8mg/L范围内呈现一定的线性关系。本文探讨了ε-PL淬灭FITC的机理,并用荧光淬灭法检测了米饭样品中ε-PL的含量。结果表明,所测得的ε-PL含量与实际添加量较为接近。
A new highly sensitive and selective fluorometric method has been developed for the determination of ε-PL using fluorescein isothiocyanate as fluorogenic reagent(λex=495 nm,λem=519 nm).In 2% ethanol medium,ε-PL can make fluorescence isothiocyanate quenched.The linear range was 0.1-0.8mg/L.In this study,the fluorescence quenching mechanism of ε-PL to FITC was discussed,and the method has been applied to the determination of ε-PL added into rice with satisfactory results.
