医用皮肤再生膜等材料生物学试验的相关性探讨 被引量:3

EXPLORATIONS ON CORRELATION OF BIOLOGICAL TESTS OF SKIN REPRODUCTIVE MEMBRANE

摘要 目的:从动物整体、细胞和分子生物的不同角度观察医用皮肤再生膜等医用生物材料的生物相容性及其相关性。方法:将医用皮肤再生膜、整形植入硅橡胶、义齿基托树脂和人工骨水泥等生物材料按ISO标准要求制备浸提液,进行一系列急性全身性毒性试验、溶血试验、细胞毒性试验以及应用分子生物技术提取细胞总RNA和细胞外基质中胶原和非胶原蛋白的mRNA基因表达等试验,综合分析试验结果及其相关性。结果:在该研究的急性全身性毒性试验和溶血试验中,所有被测生物材料都显示阴性结果。在细胞毒性试验和基因表达等分子生物学试验中,口腔印模硅橡胶表现出中等毒性以上的阳性结果。在不同浓度作用的细胞毒性试验中,随着该材料浸提液的浓度被梯度稀释,其毒性逐步减低,当浓度达原浸提液的25%时,所有被测生物材料在细胞毒性试验中都显示阴性结果。而在分子生物学的试验中,该材料浓度仅达原浸提液的10%时,即可呈现出明显的阳性结果:使细胞总RNA量减少,抑制Ⅰ型胶原、纤联蛋白的基因表达。经Spearman秩相关(STATPAL软件包)统计分析:整体试验与细胞毒性试验和基因表达试验之间不相关,细胞毒性试验和基因表达试验之间有良好的相关性。结论:该研究结果提示,对医用生物? Objective: To observe the biocompatibilities of biomaterials and discuss the correlation of different datas by cellular and molecular biological tests. Methods: The extractions of the skin reproductive membrane, the implantable silicone rubber for the plastic surgery, silicone rubber for the dental impression, risen of denture base (PMMA) and the artificial bone cement were prepared respectively. Several tests were performed, which included the acute systemic toxicity, hemolysis, cytotoxicity, quality of total cellular RNA and mRNA gene expression of the collagen and non-collagen protein.The biological compatibility and correlation were analyzed and compared synthetically from the data of the tests. Results: In the acute systemic toxicity and hemolysis, all specimens showed negative results. The tests of cytotoxicity, total cellular RNA and gene expression, silicone rubber of dental impression showed positive results that were over middle degree toxicity. Silicone rubber of dental impression could show positive result:the decreased quality of total cellular RNA and the inhibition of gene expression of type I collagen, fibronectin even at the concentration of 10% original extraction. It was found that there was no correlation between the acute systemic toxicity, hemolysis and cytotoxicity, gene expression, while the last two tests had a significant correlation by a correlated statistical analysis of Spearman's Rho (STATPAL). Conclusion: Results of these experiments suggested that evaluation on biocompatibility of biomterials should be done at the levels of the whole, cellular and molecular biological tests entirely.
