从生物材料对细胞总RNA量的影响初探其分子生物相容性 被引量:3

摘要 目的:从细胞总RNA量的变化观察生物材料有无毒性。方法:将生物材料浸提液作用于成纤维细胞,用异硫氰酸胍一步法抽提细胞总RNA,经RNA电泳和检测和计算其RNA浓度,观察细胞总RNA量的变化。结果:不同的生物材料可引起细胞总RNA量的变化,尤其某种有毒的硅橡胶使细胞总RNA量被严重抑制。结论:观测细胞总RNA量并结合细胞毒性试验及其他分子生物学试验可作为综合评价生物材料分子生物相容性的参考。 This is a very important that observation of cellular total RNA to explore toxicity of biomatcrials and compare with the result of cytotoxicity testing to estimate the biocompatibility of ilu-medical biomatcrials and the safety for human body. Method:The extracted solution of biomaterials were diluted 10 times and were co -cultured with L-929 fibroblast for 48h. Using the single -step method of RNA isolation by acid guanidinium isothiocyanate - phenol - chloroform extraction. The cellular total RNA was fraction and showed by 1.0% agarose gel containing the foramldehyde. The concentration of the cellular total RNA was measured and calculated by a DNA/RNA Calculator. Re suit:Different biomterials caused the different volume of the cellular total RNA. The reproduced mem brane of the skin,the poly methyl - methacrylate for dentistry and the implanted silicone rubber for the plastic surgery,which volume of cellular total RNA were similar with the negative control. While a silicone rubber for the oral impression of dentistry, which volume of cellular total RNA was similar with the positive control of the phenol. Conclusion:Measurement cellular total RNA of biomterials can use for explore the molecular biocompatibility of biomaterials.
