摘要
目的探讨血瘀证兔模型血管内皮细胞培养的形态结构改变及建立血瘀证细胞损伤模型的可行性。方法从兔血瘀证模型直接取材主动脉,进行内皮细胞培养,通过光镜与电镜观察原代和传代培养的内皮细胞形态结构。结果造型兔原代培养的内皮细胞融合后,光镜下发现呈多角形镶嵌状紧密排列,细胞质中可见有暗色颗粒,极少数细胞形状巨大及混杂有一定数量的平滑肌细胞。电镜检查发现胞体内饮液泡数量增多,少数细胞核形状扭曲,线粒体结构肿胀欠清晰,内质网扩张呈空泡状。且造型兔细胞生长较缓慢,至融合状态的时间较正常兔长。结论提示造型兔原代培养的血管内皮细胞出现病理性损伤,为建立血瘀证的细胞损伤模型提供了部分实验依据。
Objective:To explore morphological changes on culture of the vascular endothelial cells from Xueyu syndrome model in rabbits.Methods:Endothelial cells obtained from rabbit′s aorta with Xueyu syndrome model were cultured and identified by light microscopic,electron microscopy and immunofluorescence for rabbits antihuman Ⅷ factor serum.Light microscopic and electron microscopy observed the primary cultured and subcultured endothelial cells.Results:The primary cultured endothelial cells from Xueyu syndrome model rabbits were round to polygonal in shape,and displayed cobblestone appearance typical for endothelial cells.These cells contained dark granules in the cytoplasm.A few giant endothelial cells and smooth muscle cells were present among typical endothelial cells.Electron microscopic observation revealed that the quantities of endocellular pinocytotic vesicles increased.In a few endothelial cells,nucleusese were distorted in shape,mitochondria swollen in formation and fuzzy,endoplasmic reticula dilated to hollow vesicle in shape.Cell growth of endothelial cells from Xueyu syndrome model rabbits was slower than that of endothelial cells from normal rabbits.Conclusion:These findings suggest that there is the pathological injury occurring in primary cultured endothelial cells and also provide experimental data for the cellular model of Xueyu syndrome.
