摘要
作者用放免法测定6-Keto-PGF1a,用发色底物S_(2390)测定纤溶酶原激活物(Plasminogen activator,PA)以及它的抑制物(Plasminogen activator inhibitor,PAI)。用发色底物S_(2238)间接测定血栓调理蛋白的活性,研究了丹参和丹参素对培养的牛内皮细胞分泌上述生物活性物质的影响,及其抗凝和纤溶的机理。结果提示:丹参注射液能促进牛内皮细胞分泌PA,提高PGI_2的产生量,降低PAI的活性。人工合成的丹参素也具有同样作用。此外,丹参还能增加牛内皮细胞膜表面血栓调理蛋白的活性,而丹参素尚未观察到上述作用。
The effects of injectio Salviae Miltiorrhizae and its component 'Danshensu' on the production of PA, PAI which are related with fibrinolytic system, on PGI_2 related with anticoagulant system and on the expression of throm-bomodulin in Cultured bovine endothetial cells were studied. 6-keto-PGF1_a was measured with RIA, PA, PAI and thrombomodulin activities were measured with chromogenic substrates S_(2390) and S_(2238) respectively. The results showed that injectio Radix Salviae Miltiorrhizae could promote PA activity aud PGI_2 produced by bovine endothelial cells (BEC). It could inhibit PAI activity produced by BEC. Its component 'Danshensu' had the same effects. Beside, injectio Salviae Miltiorrhizae could increase thrombomodulin activity on the surface of BEC, whereas 'Danshensu' could not.
