赤子爱胜蚓五种纯化纤溶酶的理化性质研究 被引量:6

摘要 从赤子爱胜蚓(Eiseniafoeliad)粗制品中纯化出五种纤溶酶组分(Ⅰ、Ⅶ、Ⅷ、Ⅸ、Ⅹ),在PAGE中均呈现单一带,并富含酸性氨基酸,酪氨酸含量也较高;SDS-PAGE测得分子量分别为34000、30500、27000、24500和21500;pI为pH3.80~4.25不等;最大吸收峰均在276nm;在pH6.0~10.0及50℃以下有较高的稳定性;最适pH在8.5左右;最适温度为(60±2)℃。组分Ⅰ在70℃(10min)时活性不变,对某些变性剂表现出一定耐受性,其活性不被贵州小慈菇蛋白酶抑制。 Five fibrinolytic enzymes (Ⅰ、Ⅶ、Ⅷ、Ⅸ、Ⅹ) were isolated and purified from the earthworm (Eisenia foelied).They were single band determined by PAGE.EFE components all contain a very high proportion of acidic amino acid (Asp and Glu) and rich in tyrosine. Their molecular weight is 34 000、30 500、27 000、24 500 and 21 500 determined by SDS PAGE. The isoelectric points is between pH 3.80 and 4.25. The maximum UV peak of fibrinolytic enzymes of earthworm (EFE) is at 276 nm. They are relatively stable in the pH range of pH 6.0~10.0, below 50℃. The activity of EFE I is constant at 70℃, 10 min. EFE I has more resistance to some dena turants and is not inhibited by protease inhibitor from minor arroahead from Guizhou. The optimal pH of five EFEs is about pH 8.5 and optimal temperature is (60±2)℃.
