摘要
目的:建立一种用异硫氰酸苯酯柱前衍生的高效液相色谱方法测定金银花中17种氨基酸含量的方法,为金银花药性物质基础研究提供实验依据。方法:水解金银花总蛋白,以异硫氰酸苯酯进行柱前衍生,用HPLC测定金银花中氨基酸峰面积并计算其含量。结果:各种氨基酸均能达到基线分离。结论:高效液相色谱柱前衍生法可作为金银花药材中初生成分的测定手段之一。
Objective:To establish a method for determination of amino acids Flos Lonicerae and provide experimental basis for the physical basis of properties. Methods:The total proteins were separated,after that,hydrolyzed the total proteins,followed by pro-column derivation with Phenylisothiocyanate( PITC) and determined peak area of every amino acid by RP-HPLC method,at last calculated the cotent of every amino acid. Results:Using HPLC every amino acid can be separated well. Conclusion:This meth-od is a good way in the determination of amino acid content.
