摘要
以壳聚糖为载体,戊二醛为交联剂,制备固定化谷氨酸脱羧酶(GAD),并研究固定化反应中各个因素对固定化GAD活力的影响。结果表明,壳聚糖固定化谷氨酸脱羧酶的最佳条件为:戊二醛质量分数3%,处理时间16~24h,处理温度40℃,酶吸附温度4℃、酶吸附时间24h以上,给酶量为70.8mg/g壳聚糖。
Glutamate decarboxylase(GAD)in rice bran was immobilized with chitosan using glutaraldehyde as crosslinking agent.The influence of each factor in the immobilization was studied on the activity of immobilized GAD.The results show that the best condition of immobilization of glutamate decarboxylase with chitosan were:3% glutaraldehyde solution,processing time for 16~24h,treatment temperature is 40℃,enzyme adsorption temperature is 4 ℃,enzyme adsorption time over 24h,amount to enzyme to chitosan 70.8mg/g.
