愈痫灵颗粒对戊四氮致痫大鼠BDNF表达与认知障碍的影响 被引量:2

EFFECTS OF YUXIANLING GRANULE ON BDNF EXPRESSION AND COGNITIVE DYSFUNCTION IN PTZ-INDUCED EPILEPTIC RATS

摘要 目的:探讨愈痫灵颗粒(YXLG)对痫性大鼠海马脑源性神经营养因子(BDNF)表达与认知障碍的影响。方法:选取150只清洁级SD大鼠,从中随机选取10只作为正常组。其余动物用戊四氮(PTZ)造模。将符合模型标准的大鼠随机分为4组,即愈痫灵颗粒组、拉莫三嗪组、丙戊酸钠组和模型组。用跳台实验、开场实验测试大鼠的学习、记忆能力,采用免疫组织化学方法检测BDNF在海马内表达的变化。结果:开场实验:抗痫药物干预后,各组学习、记忆成绩比较有显著差异(P<0.01),模型组与愈痫灵颗粒组比较有显著差异(P<0.01)。跳台实验:抗痫药物干预后,各组反应期和潜伏期及学习、记忆成绩比较有显著差异(P<0.01),模型组与正常组比较有显著差异(P<0.01),愈痫灵颗粒组与正常组比较无显著差异(P>0.05)。BDNF表达:各组大鼠海马BDNF阳性细胞平均灰度值与平均光密度比较有显著差异(P<0.01),模型组与愈痫灵颗粒组比较有显著差异(P<0.01)。结论:提高PTZ致痫大鼠海马区BDNF的表达是愈痫灵颗粒改善癫痫大鼠认知功能的机制之一。 Objective:To discuss the effects of yuxianling granule on brain-derived neurotrophic factor(BDNF)expression in hippocampus of epileptic rats and cognitive dysfunction.Methods:10 rats were taken as normal control from 150 rats,the other were given pentylenetetrazol(PTZ)to build models.Rats that consistent with the standard model were divided into 4 groups,yuxianling granule,lamotrigine,sodium valproate and model groups,applying opening experiment,experimental platform to test the study and memory capacity of rats;hippocampus BDNF expression was determined by immunohistochemical method.Results:The opening experiment results showed that there was significant difference among each medicine group on study and memory capacity,there obvious difference between yuxianling granule and model groups.The results of diving platform experiment showed that there was obvious difference among each group on response and latency periods,study and memory score,but there was no difference between yuxianling group and normal group.There was significant difference among each group on the average gray value of positive cells and average optical density of BDNF expression in hippocampus.Conclusion:Enhancing PTZ-induced epileptic rats' hippocampus BDNF expression was one of the mechanism that yuxianling granule improved rats' cognitive function.
