摘要
目的:为进一步探讨膀胱肿瘤的治疗途径。方法:利用Lipofectamine将外源性野生型p53基因导入小鼠膀胱移行细胞癌BST739细胞中,并得到表达。结果:细胞在体外标准条件下,生长增殖率降低。流式细胞计检测:细胞周期分析显示G0+G1细胞比例明显增高,凋亡指数升高,细胞生长速度减低,细胞增殖能力下降,体内接种致瘤性下降。结论:增加野生型p53的基固表达能抑制恶性肿瘤的生长。
Objective: The aim of this study is to present a new therapy for bladder cancer. Meth-ods: A mouse bladder transitional cell carcinoma cell line MBST739 was transfected with wt - p53 by Lipo-fectamine. The plasmid PC - SN3 with wt - p53 - cDN Aexpressed in the mouse bladder cancer cells. Re-sults: After wt - p53 transfection, the cell growth rate decreased in vitro under standard condition. FCMexamination disclosed that the cell ratio in GO + G1 and the apoptosis index increased. The number of Ag-NORs and the carcinogenicity of cells reduced- Conclusion: The results showed that the growth of cancercells could be suppressed by wt - p53 gene.
