摘要
目的:观察自身免疫调节因子(AIRE)对CD4+CD25+Treg细胞的影响,探讨其在外周免疫耐受中的作用及机制。方法:pEGFPC3-AIRE质粒通过脂质体介导转染RAW264.7细胞;通过流式细胞术检测CD4+CD25+Treg细胞的数量;采用半定量RT-PCR法检测Foxp3和转化生长因子β(TGF-β)mRNA的表达;采用ELISA法检测TGF-β1水平。结果:AIRE转染的RAW264.7细胞可直接或间接诱导CD4+CD25+Treg细胞的产生及Foxp3mRNA表达增强(P<0.05)。结论:表达在外周抗原提呈细胞(APCs)上的AIRE可直接和(或)间接影响CD4+CD25+Treg细胞的诱导产生及功能,从而可能在外周免疫耐受中发挥重要作用。
Objective To observe the effects of autoimmune regulator (AIRE)on CD4+CD25+Treg cells,and explore the functions and mechanisms of AIRE in peripheral tolerance.Methods RAW264.7cells were transfected with pEGFPC3-AIRE plasmid via lipofectin.Flow cytometry was used to determine the amount of CD4+CD25+ Treg cells.Then semi-quantitative RT-PCR was employed to detect the expressions of Foxp3and TGF-β1mRNA. The level of TGF-β1was detected with ELISA kit.Results AIRE transfected RAW264.7cells could directly or indirectly induce the generation of CD4+ CD25+ Treg cells and the increased expression of Foxp3 mRNA. Conclusion The AIRE expressed in the peripheral antigen presenting cells(APCs)could directly and/or indirectly induce the generation and functions of CD4+CD25+Treg cells,and probably plays a crucial role in the peripheral immune tolerance.
