摘要
目的探索应用胃泌素受体拮抗剂调控胃癌细胞自分泌生长的作用及其机制。方法建立胃癌细胞株ＭＫＮ４５移植瘤的动物模型，观察胃泌素受体拮抗剂丙谷胺对移植瘤的面积、重量、胃癌细胞ＤＮＡ及血清促胃液素含量的影响。结果第１６天丙谷胺组移植瘤面积（６１．４１±１８．５７）ｍｍ２较对照组（９４．８６±２８．０５）ｍｍ２明显缩小（Ｐ＜０．０５），第２８天时差别更明显（１２５．５７±４０．２６）∶（２１１．５９±５０．０８）ｍｍ２。第２８天时丙谷胺组及对照组的移植瘤重量分别为（８４７．９±６９．６）ｍｇ及（１１７７．７±８３．５）ｍｇ（Ｐ＜０．０５）。丙谷胺组胃癌细胞ＤＮＡ指数、平均ＤＮＡ含量及Ｓ期细胞比例均低于对照组。丙谷胺对血清促胃液素含量没有影响。结论丙谷胺对胃癌细胞株ＭＫＮ４５移植瘤有抑制生长的作用；丙谷胺发挥抗肿瘤作用是通过改变胃癌细胞ＤＮＡ代谢过程，而与血清促胃液素含量的改变无关。
Purpose To study the effect and mechanism of gastrin receptor antagonist on the regulation of the autocrine growth of gastric cancer cells. Methods The animal model of xenotransplanted gastric cancer cells MKN 45 was established,and the effect of a gastrin receptor (GR) antagonist,proglumide,on xenotransplanted tumor area, tumor weight, tumor cells DNA and the serum level of gastrin was investigated . Results Tumor area was smaller in the proglumide group (61.41±18.57)mm 2 than that in the control group (94.86±28.05)mm 2 after 16 days treatment ( P <0.05).Tumor area became much smaller after 28 days treatment in the proglumide group than that in the control group (125.57±40.26)mm 2 vs (211.59±50.08)mm 2.Tumor weight was (847.9±69.6)mg and (1 177.7±183.5)mg in the proglumide group and the control group respectively after 28 days treatment ( P <0.05).It was found that DNA index,average DNA contents and S phase fraction of gastric cancer cells in the proglumide group were lower than those in the control group.Proglumide did not affect the serum level of gastrin. Conclusions Proglumide plays an inhibitory role in the growth of xenotransplanted gastric cancer cells MKN 45 .The inhibitory process of proglumide is by altering the DNA metablism of gastric cancer cells and not related to the serum level of gastrin.
