液体悬浮培养产发菜多糖的质量分析 被引量:3

Quality Analysis of Nostoc flagelliforme Polysaccharide Produced by Liquid Suspension Culture

摘要 从性状、多糖含量、pH值、水分含量、灰分含量、粘度、澄清度、重金属含量和微生物检验方面对液体悬浮培养发菜粗多糖进行质量分析。结果表明,浓度为100mg/L的发菜粗多糖溶液其多糖含量为43.19±1.00mg/L;性状为白色或淡黄色无味粉末,其溶液浓度在0.1~4.0mg/mL时,pH值范围在6.0~7.0之间,是一种酸性多糖;水分含量为10.93±0.10%,灰分含量为4.92±0.05%;浓度为1%的多糖溶液粘度为22.6mPa·s(20±1℃,200r/min);多糖浓度为0.05mg/mL时,溶液澄清;重金属含量(mg/kg)分别为Pb:0.83±0.08,Hg:0.13±0.03,As:0.44±0.06,Cd:0.063±0.001;浓度为1.0mg/mL的溶液中细菌总数为2cfu/mL,大肠菌群数小于0.03MPN/mL,霉菌和酵母数均小于1cfu/mL,志贺氏菌、金黄色葡萄球菌和沙门氏菌等致病菌均未检出。 Nostoc flagelliforme was liquid-suspension cultured for production of the Nostoc flagelliforme polysaccharide and its quality was characterized by determination of the polysaccharide content,appearance,pH value,moisture,ash,viscosity,clarity,heavy metal contents and microbial assay.The results showed that the polysaccharide content was 43.19±1.00 mg/L with its concentration of 100 mg/L.The polysaccharide was a white or pale yellow odorless solid.The pH value of this acidic polysaccharide was between 6.0 and 7.0 when its concentration between 0.1 and 4.0 mg/mL.The water and ash contents of the polysaccharide powder were 10.93±0.10% and 4.92±0.05%,respectively.The viscosity of the polysaccharide solution was 22.6 mPa·s(1%,20±1 °C,200 r/min) with a concentration of 1%.And the solution was clear at 0.05 mg/mL.Atomic absorption spectrophotometry showed its heavy metal contents(mg/kg) were as follows:Pb,0.83±0.08;Hg,0.13±0.03;As,0.44±0.06;Cd,0.063±0.001.The bacterial amount was 2 cfu/mL and the number of Coliforms was lower than 0.03 MPN/mL in 1.0 mg/mL of the polysaccharide.Both the numbers of mold and yeast were lower than 1.0 cfu/mL.In addition,Shigella,Staphylococcus aureus and Salmonella were not detected in the achieved Nostoc flagelliforme polysaccharide.
