摘要
目的为了提高染料木素的生物利用度,制备染料木素磷脂复合物,并对其光谱性质进行研究。方法以染料木素与大豆卵磷脂的结合率为评价标准,对反应溶剂、反应时间、反应物浓度、反应温度和投料比5个因素对结合率的影响进行了考查,利用UV、IR、XRD、DSC等分析技术对复合物结构进行分析。结果最佳制备条件为:四氢呋喃为反应溶剂,染料木素质量浓度为2 mg.mL-1,投料摩尔比为1∶3,反应时间为3 h,反应温度为50℃,该条件下产物结合率达95.4%。结论磷脂复合物结构不同于单体及物理混合物,染料木素与卵磷脂极性部位经范德华力、氢键等作用结合形成了复合物。
OBJECTIVE To investigate the preparation and action spectrum properties of genistein phospholipid complex.METHODS The combining ratio of genistein and phospholipid was regard as the assessment standard.Five main factors including reaction solvent,reaction time,reactant concentration,reaction temperature and the ratio of reactants on this reaction were investigated.Then,the the action spectrum properties of genistein-phospholipid complex including UV spectrum,IR spectrum,differential scanning and X-ray diffraction spectra were analyzed.RESULTS The optimum conditions were as follows: reaction solvent was tetrahydrofuran,the concentration of genistein was 2 mg·mL-1,the ratio of reactants was 1∶3,reaction time was 3 h,reaction temperature was 50 ℃.In optimum conditions,the yield was above 95.4%.CONCLUSION The structure of genistein-phospholipid complex had some difference between reactant monomer and physical mixture of genistein and phospholipid.The formation of the complex is based on the van der Waals forces and hydrogen bond between phospholipid polar group rounding phosphorus atom and genistein.
