RP-HPLC双波长同时测定关节止痛膏中硫酸阿托品、盐酸苯海拉明、辣椒碱和二氢辣椒碱的含量 被引量:6

Simultaneously determination of atropine sulphate,diphenhydramine hydrochloride,capsaicin and dihydrocapsaicin in pain-relieving plaster for arthritis by RP-HPLC

摘要 目的:建立用高效液相色谱同时测定关节止痛膏中主要药效成分硫酸阿托品、盐酸苯海拉明、辣椒碱及二氢辣椒碱的含量。方法:采用RP-HPLC双波长法,色谱柱Alltima C18(4.6 mm×250 mm,5μm);流动相乙腈-0.1%磷酸,梯度洗脱程序;体积流量1 mL·min^-1;检测波长210,280 nm;柱温30℃。结果:硫酸阿托品在2.0150.25 mg·L^-1,盐酸苯海拉明在15.08377.00 mg·L^-1,辣椒碱在5.02125.50 mg·L^-1,二氢辣椒碱在5.03125.75 mg·L^-1与峰面积呈良好的线性关系。加样回收率分别为99.00%(RSD 0.95%),99.89%(RSD 1.2%),100.13%(RSD 1.5%),99.51%(RSD 1.4%)。结论:此方法准确、快速分析关节止痛膏中4种主要成分,灵敏度高,重复性好,可控制关节止痛膏的质量。 Objective: To establish RP-HPLC method for determination of atropine sulphate,diphenhydramine hydrochloride,capsaicin and dihydrocapsaicin in pain-relieving plaster for arthritis.Method: The sample were separated on an Alltima C18 Column(4.6 mm×250 mm,5 μm)with the moblie phase of CH3CN-0.1%H3PO4.Flow rate was 1 mL·min^-1The detective wavelength was set at 210 and 280 nm.Column temperature was 30 ℃.Result: The calibration curve for atropine sulphate,diphenhydramine hydrochloride,capsaicin and dihydrocapsaicin revealed linearity in the range of 2.01-50.25,15.08-377.00,5.02-125.50,5.03-125.75 mg·L^-1,respectively.The recoveries of atropine sulphate,diphenhydramine hydrochloride,capsaicin and dihydrocapsaicin were 99.00% with RSD of 0.95%,99.89% with RSD of 1.2%,100.1% with RSD of 1.5% and 99.51%,with RSD of 1.4%,respectively.Conclusion: The method is simple,rapid and accurate,which is suitable for the quality control of pain-relieving plaster for arthritis.
