The human tumor suppressor
gene nm23-H1 was cloned into retroviral vector pLXSN and constructed an expressing vector
pLXSN-nmh1. This recombinant plasmid was transfected into ψCRIP paking cell line by
calcium phosphate precipitation technique. The colonies capable of secreting product of
nm23-H1 gene and the recombinant retroviruses were selected by G418. The integration and
expression of nm23-H1 gene were detected by PCR and Western blot hybrization
respectively. This study demonstrates a crucial role of retroviruses as gene therapy and
transgenosis.
