摘要
使用丹黄酰氯对目标生物胺进行柱前衍生,采用WATERS Atlantis dC18色谱柱(4.6×150mm ID,5μm),以乙腈和水作为流动相,进行梯度洗脱,流速为0.6mL/min,采用UV紫外检测器,柱温为30℃,检测波长254nm,对8种生物胺进行分离分析。结果显示,回收率超过88%,最低检测限为0.01mg/L~0.1mg/L,保留时间和峰面积相对标准差低于2%。
The dansyl chloride was choose as target biogenic amine for pre column derivatization. Using WATERS Atlantis dC18 chromatographic column (4.6×150mm ID, 5um), 8 biogenic amines were separated and analyzed by RP-HPLC. The chromatographic conditions of RP-HPLC were as fol- lows: the mobile phase for gradient elution consisted of acetonitrile and water, the flow rate was 0.6ml/min, the UV detector was used, the column temperature was 30℃ and the detection wavelength was 254nm. The results showed that the recycle rate was higher than 88%, the minimum detection limit was 0.01-0. 1 mg/L and the retention time and peak area were 2% lower than the standard deviation.
