摘要
目的:研究γ-干扰素(IFN-γ)及白介素-4(IL-4)等细胞因子在流产过程中发挥的作用及其机制.方法:实验孕鼠随机分为正常孕组、流产模型组、流产模型+IFN-γ抗体组.均于孕12d剖腹观察妊娠结局并取材,酶联免疫吸附法测定各组大鼠血清IFN-γ、 IL-4水平,免疫组织化学技术检测卵巢黄体IFN-γ、 IL-4蛋白表达情况.结果:血清测定数据显示,流产模型组IFN-γ水平高于正常孕组,IL-4水平低于正常孕组;流产模型+IFN-γ抗体组IFN-γ水平低于流产模型组,IL-4水平高于流产模型硎组.免疫组织化学阳性细胞光密度值变化趋势与血清测定结果基本一致.结论:IFN-γ、 IL-4与流产具有相关性,大鼠血清与卵巢黄体中两种细胞因子表达的改变可能是流产发生的重要因素之一.
Objective: To study the effect and mechanism of cytokine of IFN-γ and IL-4 in abortion. Methods: Pregnant rats were randomly divided into experimental normal pregnant, abortion model and abortion model+ IFN-y antibody. We cut the belly to observe the outcome on the 12th day in all groups. Enzyme-linked immuno-assay was used to assay the concentration of IFN-γ and IL-4 in blood serum. Immunohistochemistry were used to detect expression of IFN-γ and IL-4 protein in corpus luteum of ovary. Results: The serum IFN-γ level of the model group was markedly higher than that in the control group and IL-4 level was lower than that in the control group. The serum IFN-γ level was markedly lower than that in the model group and IL-4 level was higher than that in the model group. Conclusion: IFN-γ and IL-4 may be related to abortion. The expression change of two cytokine in serum and corpus luteum of the ovary is important to the mechanism of abortion.
