摘要
采用活化剂碳二亚胺(EDC)和N-羟基琥珀酰亚胺(NHS)对邻巯基苯甲酸(MBA)自组装修饰的石英晶体金电极表面进行活化,再将三唑磷单克隆抗体共价固定于电极表面,建立了一种用于快速检测农药残留的压电免疫生物传感器。采用浸入-干燥法和流动注射法检测目标农药并对实验过程进行分析。两种方法均可在40~60min内直接检测目标农药,浸入-干燥法的检出限为0.04mg/L,流动注射法的检出限为1.0mg/L。使用8mol/L尿素成功解离了抗体-抗原免疫复合物,实现了免疫传感器的再生。此传感器的选择性好,可以重复使用。
A piezoelectric immunosensor was developed for rapid detection of triazophos.It was based on the immobilization of triazophos monoclonal antibodies onto a monolayer of 2-mercaptobenzoic acid (MBA) self-assembled on an AT-cut quartz crystal's Au electrode surface with N-hydroxysuccinimide (NHS) ester as a reactive intermediate.Two analytical procedures,dip-and-dry and flow-through methods,were investigated.The immunosensor could detect the target pesticide within 40-60 min,the detection limit of triazophos was 0.04 mg/L for dip-and-dry method and 1 mg/L for flow injection method.The repeated use of immunosensor was possible by regeneration using 8 mol/L urea.
