摘要
目的探讨放射性核素标记重组改良型人膜联蛋白V(His-AnnexinV)的方法,并应用于体内显像的研究。方法取不同剂量的His-AnnexinV以生理盐水溶解,加入缓冲液,再分别加入不同剂量的还原剂SnCl2,然后在其中分别加入不同活度的99mTcO4-进行直接标记,混匀,条件均为常温下震荡,37°孵化,取出自然冷却到室温。结果 His-AnnexinV500μg,加入40μg还原剂SnCl2,然后加入185~296MBq99mTcO4-进行直接标记,标记率可达到95%以上;99mTc-His-AnnexinV在动脉粥样硬化兔模型中,可有效检测动脉粥样硬化斑块。结论 99mTc直接标记His-AnnexinV,标记率高,并可成为动脉粥样硬化斑块的显像剂。
ObjectiveTo investigate labeling methods of 99mTc-His-Annexin V,and to apply this method to imaging research in vivo.MethodsTo get different dose His-AnnexinV,buffer solution and then different doses of SnCl2 were added.And solution was mixed with 99mTcO4-of different activity for direct labeling.and then shaken in common temperature,incubated in 37℃ and natural cooled at room temperature.ResultsA total of 500μg 99mTc-His-AnnexinV were added with 40μg SnCl2 add 185~370 MBq 99mTcO4 for direct labeling,the labeling rate arrived at 95%.And 99mTc-His-AnnexinV could effectively detect arthrosclerosis plaque for rabbit arthrosclerosis model.ConclusionThe labeling rate of 9mTc-His-Annehin V is high by direct method,and it can become a kind of noninvasive method for detecting atherosclerotic plaque.
