高效液相色谱法测定5′-三磷酸腺苷含量 被引量:3

摘要 采用高效液相色谱法测定5′-三磷酸腺苷含量。色谱条件为:XTerraTMRP18(3.9 mm×150 mm,5μm)色谱柱,流动相为0.05 mol.L-1pH值7.0的NH4H2PO4缓冲溶液,检测波长259 nm,流速1.0 mL.min-1,柱温(25±2)℃,进样量20μL。在10~80 mg.L-1范围内,ATP浓度与其259 nm处吸光度值呈良好线性关系,R=0.9994;检测重复性较好,平均回收率为98.87%,整个色谱过程可在7 min内完成。该方法也可用于ADP和AMP的分离检测。 A HPLC method had been developed to determine the content of ATP(Adenosine triphosphate).The chromatographic conditions were selected as follows:XTerraTMRP18(3.9 mm×150 mm,5 μm) column;the mobile phase was NH4H2PO4 buffer solution(0.05 mol·L-1,pH value 7.0);detection wavelength was 259 nm;the flow rate was 1.0 mL·min-1;the injection volume was 20 μL;the column temperature was(25±2)℃.The method had good repeatability and had a good linearity within the range of 10~80 mg·L-1(R=0.9994).The average recovery was 98.87%.The process could be completed within 7 min.The method was reproducible for determination of ADP and AMP.
