新疆锁阳线粒体nad 1基因第2内含子序列居群间变异分析

摘要 目的:对分布于新疆的锁阳Cynomorium songaricum 4个居群22个个体进行了线粒体nad1基因第2内含子序列的比较研究,分析序列居群差异。方法:提取锁阳DNA,用通用引物扩增线粒体nad1基因第2内含子序列,Clustal W软件比较分析序列。结果:锁阳22个样本的nad 1基因第2内含子序列变异均表现为插入和缺失,无碱基替换发生,有13个个体序列完全一致,9个个体有变异位点,变异位点为10个;4个居群间分化不明显,其中b居群可能存在一定的分化,有两个个体y4和a19存在较大的变异,可能具有一定的遗传学意义。结论:线粒体nad 1基因第2内含子序列给出的锁阳4个居群分化不明显。 Objective: To examine population variation of mitochondrial nadl intron 2 within 22 individual samples from 4 populations of cynomorium songaricum in Xinjiang. Methods: Total DNA was extracted, then the mitochondrial nadl intron 2 of samples were PCR amplified and sequenced. Sequences alignment were performed using Clustal W software package. Results: The variation of mitochondrial nadl intron 2 sequences among twenty-two individuals is base insertions or deletions, the sequences of 13 individuals are completely the same, the rest 9 samples have 10 mutation sites in total. Among the 4 populations only population b may have differentiation in the sequence, in which the variation of y 4 and a 19 sample is significant. Conclusion: The sequecnce differentiation of nad 1 intron 2 in cynomorium songaricum isn't obvious among 4 populations.
