雌激素与人尿酸盐转运子(hUAT)基因表达的相关性研究 被引量:26

摘要 目的观察不同浓度雌激素对肾小管上皮细胞(HK-2细胞)人尿酸盐转运子(hUAT)基因表达的调节作用。方法根据培养液中所含雌激素浓度由低到高,将HK-2细胞分为5组,每组均培养6瓶细胞。上述细胞在不同培养液中分别培养48h。采用荧光定量PCR法检测HK-2细胞中hUAT mRNA的相对表达量。结果所有标本均能检测到hUAT mRNA的表达,其中,与B0组(乙醇对照组)比较,B3(10-8mol/L)、B4(10-6mol/L)组hUAT mRNA水平明显升高(P<0.05),B1(10-12mol/L)、B2(10-8mol/L)组hUAT mRNA水平变化不明显(P>0.05)。与B1、B2组比较,B3、B4组hUAT mRNA水平明显升高(P<0.05)。B3组与B4组比较差异无统计学意义(P>0.05)。B1组与B2组比较差异无统计学意义(P>0.05)。B4组hUAT mR-NA水平最高,是B0组的1.87倍。结论雌二醇可上调hUAT mRNA的表达,且可能存在相应的作用阈值。 Objective To observe the effect of estradiol in different concentrations on the expression of human urate transporter(hUAT) gene in HK-2 cells.Methods HK-2 cells were divided into control and other 4 groups which were cultured with different concentrations of estradiol in each group,6 bottles of the cells were cultured in vitro for 48 hours.Then the relative quantity of hUAT mRNA in the HK-2 cells was detected by real-time fluorescent quantitative PCR assay.Results hUAT mRNA could be detected in all the samples.And the level of hUAT mRNA in the control group was significantly lower than that in the groups of B3 and B4.The quantity of hUAT mRNA in the B4 group was 187% of that in the control.Conclusion Estradiol can up-regulate the expression of hUAT gene,and there may be a Threshold.
